Before using, the culture dish have to be cleaned and disinfected. It has a significant impact on the work and can affect the acidity and alkalinity of the culture medium, depending on whether it is clean or not. Certain chemicals can inhibit bacterial growth if they are present.
Newly purchased culture dishes should be rinsed with hot water first, then soaked in a 1% or 2% mass fraction hydrochloric acid solution for several hours to remove free alkaline substances, and finally rinsed twice with distilled water.
To cultivate bacteria, sterilize with high-pressure steam (typically 6.8 * 10 to the 5th power Pa high-pressure steam) at 120 ° C for 30 minutes, dry at room temperature, or sterilize with dry heat. This entails placing the cultured blood in an oven for 2 hours at a temperature of around 120 ° C to kill the bacterial cells.
Inoculation and cultivation can only take place in sterilized petri dishes.
